vacancies in this context means "เวลาว่าง".

"do you have any vacancies for tonight?" - คืนนี้คุณพอมีเวลาว่างบ้างไหม

Khun Nan,

I believe, although it's not 100% clear, that the OP is referring to a vacancy of a room (in a hotel, perhaps) rather than the free time of a person.

If that is the case, we could say คืนนี้คุณมีห้องว่างบ้างไหมครับ

Literally, it means "enough" - to have something enough for some purpose. When it's added in front of the sentence or after the subject, it would sound politer if you want to ask for something.

คุณพอมีเวลาว่างบ้างไหม ผมอยากคุยด้วย (enough time for us to talk )

คุณพอมีเงินบ้างไหม ผมอยากจะขอยืมสักหน่อย (enough money for me to borrow)

พอจะบอกได้ไหมว่าเขาไปไหน (enough information for you to tell )
